MS2 : In-gel (In-solution) digestion and protein identification by automatic LC-MS/MS

Test Introduction

Our staff has been providing protein identification services for over eight years in Korea and overseas. Proteins’ identification are either analyzed by MALDI-TOF/TOF-MS, which we have at our facility. The latter also identifies several proteins within a sample 1D gel band or 2DE spot with in-gel digestion using protease ( such as trypsin, Endoproteinase Asp-N,,,,,.). The tryptic digest is then analyzed by LC-MS/MS. Custom-made chromatographic columns are used for desalting and concentration of the peptide mixture prior to mass spectrometric analysis. For high sensitivity applications proteolytic digests from electrophoretically separated proteins must be desalted prior to analysis. This is a very important step for mass spectrometry, because protein band/spot with lower amounts need concentrations above 50 times. The detection of gel based protein identification is about 50ng. It’s available in both silver and coomassie stained gels.

Our protein identification service includes In-gel digestion, desalting and concentration, data acquisition, and database searching. Analysis takes three business days to complete after sample arrival, and the results are e-mailed to the customer in Excel and powerpoint format.
